With a rifle you need to roll to see if you hit, then roll for damage. How does it work with grenade launchers? Do you roll the RC first? And if so what are the modifiers for no line of sight?

You roll the RC as normal. If you hit you roll a d6. On a roll of 1-2 the grenade hits on top of the spot you designated (grenade launcher doesn't target models but a spot). If the die shows a 3-6 roll another d6 and a scatter die. The grenade scatters that many inches in the direction of the scatter and hits there.
If you miss you roll 2d6 and a scatter die. Pick the highest number - the grenade scatters that many inches in the direction of the scatter die.
There are no modifiers for no line of sight.
